A metal straw and scalpel to move the pieces. link thanks! Still had a little bit of an issue with one piece.
pouring your own is alot more bullshit than you think it will be, but check this link. this is what i got. it looks legit, is PDA. PREPOURED!
good luck, have fun.
Appreciate the link. The hole seems rather small, but that's the concept we need.
I took your advice on the olive stuffer and found this one. Seems like the most useful for punching and transferring. My only problem is trying to justify spending that much on something I don't necessarily need.
Thing is I think this just looks like one of these tools.
you shouldn't need to sterilize the air but I'm sure its not hurting.
Im assuming you're pressuring cooking your agar and stuff. I use these bags to pc my tools, it probably isn't your problem, but having fresh tools is 1 less way to get contam.
I usually flame sterilize my needles, if they're fresh it's probably not the issue, but just wiping with iso doesn't always make me feel safe.
The contam in your plates look like they came with the liquid culture imo, it is growing with the mycelium. It is possible the LC is bad, I'd suggest making transfers to clean plates and trying to get something to work with. Not sure your budget but you could always buy another LC or clean plate from a good source.
I cut all my parafilm in my bed room and haven't had an issue yet. Wet plates are hard to parafilm so I personally wouldn't do that. Parafilm won't introduce contam unless your plates are very wet.
Remember when working in a flow hood the direction of flow, you don't want to have anything upstream of what you're working on. When you open plates, open them into the flow. Every move you make think about how the airflow is going, you should never have anything in front of anything else.
If I was you I'd take each plate you have and transfer to another 2 plates from each one. Good practice, and hopefully you end up with at least one that's clean. In the meantime it might be time to start pouring your own plates and maybe order another syringe.
Some sort of epigenetic evolution couldn’t happen? Or sort of a crossing with resistant strain? Maybe activate an old genetic or something; look i activated some yeast genetic into perfectly healthy myc, making it looks like yeast, yup yup just some mutation … 🙄
Rowenta Pure Air Connect Purifier https://www.amazon.fr/dp/B07BCNKFFV?ref=ppx_pop_mob_ap_share I use this, i let it turn in the room (small room 1square metter maybe) about 20/30min before starting to work. But for now i let it turned while working, you recommend to do not? I think if i shut it off and spray some water within the air i won’t have to wait 1 or 2 hours before transfer, am i right ?

I take a 1/2 inch drill bit, flip it upside down, heat up the back side of the bit and then push it through the top of the lid. Cut out a quarter sized circle from a synthetic filter disc and adhere it on top of the lid via RTV silicone.

I don't really recommend it, though. It's sort of lopsided. Makes pouring dishes a problem if you have OCD about perfectly even dishes.
