I know a few people are saying Balsamiq, but I looked at loads of wireframing and prototyping webapps (including Balsamiq), and HotGloo came out on top (in almost every way)...
One cool feature is that it has interactions, so you can emulate basic functionality (like links between pages/wireframes etc). They also have a really large library of UI elements.
