You can try running an app on the Quest side to enable wireless ADB (i.e. https://play.google.com/store/apps/details?id=com.ttxapps.wifiadb&hl=en). Without enabling it at every boot, it will not be possible either way, however.
Has anyone else had ADB over Wifi break since upgrading to Marshmallow? This is especially annoying since the USB C-to-A cable included with my Nexus 6P is too short to reach my computer. I have tried with WiFi ADB as well as Trickster MOD, and both times this happens:
adb connect 192.168.0.xxx * daemon not running. starting it now on port 5037 * * daemon started successfully * unable to connect to 192.168.0.xxx:5555
The same happens when I do adb connect 192.168.0.xxx:5555. This happens on both my Nexus 6P, stock MDB08K rooted with SuperSU 2.52 and Nexus 5, stock MRA58K+elementalX kernel rooted with SuperSU 2.52.

Another option is using an Android phone or tablet to connect instead of a PC (so you don't even need to download the tools). This WiFi ADB app works fine https://play.google.com/store/apps/details?id=com.ttxapps.wifiadb
Could a problem specific to your device. You have root right? Try ADB WiFi then use the app I mentioned earlier (Remote ADB). Run this line to enable Developer settings:
settings put global development_settings_enabled 1
Think it's built into Android 11 now.
But on older devices, if you have root, you can do it through 3rd-party apps. Been a while since I've been rooted so my memory is a bit fuzzy, but I think this app can do it on boot if you whitelist an SSID.

Does this let me use my phone as if it were usb plugged into my computer, but over wifi instead? If so that would be amazing. For some reason my Droid Turbo won't connect to adb.

EDIT: It turns out the WiFi ADB app in the How To requires root, so I'm not sure if this is possible without it.
You can use ADB over Network or check out this How To first, then you can use either Custom Quick Settings or QuickTask to create a tile. I might set this up myself.
