well you can't have a USB connection because the port is taken by the GearVR but if you use a wireless ADB connection like this https://play.google.com/store/apps/details?id=za.co.henry.hsu.adbwirelessbyhenry&hl=en it should work...
I use the Build and Run option in Unity to send the build straight to the phone. I also use an app called ADB Wireless that makes it easy to setup a wireless link, too. It will send the build over wifi instead of plugging and unpluggin the phone USB constantly. Just set up the wireless link once per session and you can leave the phone in the Gear and just send the new builds whenever you want.
The non-stereo splash screen is a problem that seems to have no solution right now. Doesn't happen to everyone and was supposed to be fixed during previous Unity updates. Happens to me and I pretty much gave up trying to figure it out for now. Relevant thread on Oculus forum.
You don't need root. All you need to do this is ADB (Android Debug Bridge).
ADB has two ways to connect: USB or over WiFi. Since you don't have a PC, WiFi it is then.
However, you may need a PC to activate ADB over WiFi, but there's an app that claims it can do it without a PC (Play Store link). You need to install this on your shield. However, the instructions on the app is misleading. In my case, I don't need to connect my device to a PC through USB to enable wireless adb, I simply clicked the large "Show my ip" button in the app and it turns on wireless adb for me.
Then, on your phone, install this (scroll down to see the download), a scrcpy port on Android. Type in the ip of your shield (should be displayed on the screen, make sure your devices are on the same network). Don't forget to enable the Nav bar option. And, voila! You should see your shield's screen on your phone and able to simulate touch.
But I've heard that the Android port of scrcpy is having some issues with landscape devices, so, if you want the best experience, you'd better get a PC and run scrcpy on it. I can provide instructions if you are interested in that.
kinda late, but have you tried wireless adb? I've used it with vysor and it's worked, since vysor connects via adb

Pada dasarnya ADB itu jalan via network, bodo amat networknya kabel/wifi (or even internet...)
Kalo lom root, tiap first connect masih pake kabel lalu bisa sesssion panjang di wireless. Benere g perlu pake app, via cmd juga bisa tapi lebih simple aja ngecek IP nya.
Kalo dah root, konek sekali pake kabel, seterusnya selamanya bisa wireless.
