Install SwipeUp Utility and, if using Nova, follow these steps. For those not using Nova, you can substitute any automation app to get the screen lock action.
I'm completely fine with no app drawer as long as there's a way to hide apps from your home screen and a quick way to search for apps. One of my favorite features in Nova Launcher is its App Search shortcut. I don't use Google Now so I have Now on Tap replaced with App Search using Swipeup Utility. This way, from any screen I can press and hold the home button for half a second to start typing in the search bar to search for apps on my phone or on Google Play. It really changed how I use my phone and I've only opened the app drawer by accident since then.

Swipeup Utility allows you to replace the assistant shortcut with a custom action (toggling flashlight, pulling down the notification panel and so on). I use it to launch Sesame Shortcuts search panel

yup, I used to use App Swap (similar to App Dialer) but moved to Sesame launched via Swipeup Utility. I love being able to open apps and search the web from the same screen.

I installed Swipeup Utility intending to get a swipe up gesture to launch the BB Hub, but I guess on Marshmallow the swipe up gesture was replaced with a long press of the home button. Normally, this would launch Google Now On Tap, but I've switched it to launch the BB Hub.
The BB keyboard, plus BBM and the BB Hub is really all I needed.
Swipeup Utility (https://play.google.com/store/apps/details?id=com.AdrianCampos.swipeuputility) lets you do something else instead of launching Google Now. One of the options is a quick Google search. (It's a long-press on Home for me, not a swipe-up, but I think this is what you mean.)
I've been using Swipe Up Utility to open the camera in video record mode. You never know when you're going to want to capture something interesting. The really nice thing about SUU is that you can configure it to just about anything you want.
I'm not sure if this is what you're talking about? This app works even on unrooted stock phones: Swipeup Utility. (I haven't tested on Lollipop personally) It allows you to change, disable, or add behaviors as well as change icons of the swipe-up function. So, it should work with any other ROM or stock.
Set SwipeUp Utility as your assistant app and then have SwipeUp launch Sesame Search by default.

For a file manager without ads, download F-Droid (an open source app service -- like an app store). There are a few options. I like the one from Cyanogenmod.
