You should install the unofficial patch, as far as I'm aware the vanilla game actually isn't finishable without it. Comes in two varieties - a patch to fix bugs and render the game playable, and the "patch plus" version that includes a bunch of lost and unfinished content.
Buy it on GOG, it's on sale there too. That version has the Unofficial Patch already installed, and you are ready to go. You don't need anything else the first time. Maybe the eye replacer mod for a purely cosmetic change.

"How much of a big deal is the vampire clan you pick? Does it greatly affect story or gameplay?" Only Nosferatu and Malkavian alter gameplay in drastic ways. Nosferatu mostly affects gameplay, since your hideous (awesome) appearance doesn't allow you to walk streets and you have to hide and navigate through sewers mostly. Malkavian twists ingame conversations to resemble "madman speak", so it's a bit harder to get what they mean, though certainly not impossible. The rest are played more conventionally.
"Are you locked into a single clan? Can you multi-class/clan?" Yes. Pick one at the start and you will be playing it for the rest of the game.
"Can you switch clans, or use abilities from other clans?" You can't switch clans, but some of them share the same abilities. For example, both Malks and Nosferatu have Obfuscate (extra stealth).
"And if the answer is no to most of the above, is this a popular repayable game? Like, is it commonplace to do a replay, one for each clan?" It certainly has replay value. Pick a different clan and you will be playing the game differently. I recommend to pick a conventional clan for your first playthrough.

It lets you choose from 2 versions when you install it. Plus version and normal version. Normal version are just bug fixes and plus version adds and re balances few things.

It's pretty straightforward.
The patch will place a shortcut on your Desktop called "VTM Bloodlines Unofficial Patch." Launch the game through that shortcut to play the patched version. Alternately, right-click your game in your Steam library and click Properties, Set Launch Options, type -game Unofficial_Patch and click OK. I recommend this second method. I've found that if you use the Desktop shortcut Steam doesn't monitor your playtime and you miss out on a few things like the ability to use alt-tab, etc.
The basic version of the unofficial patch comes pre installed on GOG. If you want to run the Plus version of the patch after a play through or two you can download the patch here then just follow the directions on the installer.
If you're considering getting it at some point. GOG is the best option. It's easier yo mod that one and it comes with as at least the some I am righting this. The latest basic version of the Unofficial Patch. If you want the plus content you need the exe for it. Which you get can get here.

The current stable version of the Unofficial Patch us 10.1. 10.2's release is coming up though. But if you start a character on 10.1 do not carry the character over to the new version. If you do it most likely will introduce bugs.

I used AvP2 queen sounds to make the deathclaw more fearsome, the VtMB part are the claw sounds which are enhanced with the katana slash and hit sounds. All custom remixed with the original sounds.

It's just creating that .bat which does the job of the Windows shortcut that we cannot run. The wineskin which contains Steam and the game is then set up so that when you double click its icon, it launches the .bat file. There's also a technique to be able to run other .exe's, like the patch installer for example.

Most unique out of the two is Nosferatu. It changes how you play the game and characters react differently to you; you can even kill someone early in the game (at the cost of Humanity) by repeatedly talking to said person. You do get Masquerade violations if you're seen by people, but it's a bit exaggerated; you can still run on the streets (though Chinatown is a different story) and peds will run away if they see you. Stay close to them and then that's when you get a Masquerade violation. Just have to hurry and move. At least, that's from my experience.
The only unique thing about Tremere is their Thaumaturgy discipline; only one character has a unique reaction towards the Tremere PC and that's a Plus patch exclusive, I believe. Although Tremere has the best haven in the game, it just sucks it takes a bit of time to get (and you can easily fuck it up, too).
Combat-wise, I prefer Tremere; Thaumaturgy is fun and makes it easier. Although, Obfuscate discipline is another favorite of mine.

The Clan Quest mod is the one total overhaul mod that I will always recommend, even on a first-time playthrough. It's essentially an expansion pack at this point because of the vast amount of extra content it offers and it incorporates a lot of the content from the plus patch into the game anyway. I believe CCM 4.1 uses the Unofficial Patch 9.6. Combine that with the X20 Mod and I think that you pretty much get the definitive version of the game at that point.
The Clan Quest mod is phenomenal.
Ok I'm super new to vtmb as well as computer gaming (having bought a laptop solely to play this game before bloodlines 2 lol) but I managed to figure it all out;

This part's important and took me a couple tries to figure out. When you download the patch, you must either select the box that says patch plus if youd like to run with patch plus OR not check the box to run with just the 10.4 patch. Then install it. Its pretty straight forward.
Finally, to make the patch run in the steam client, right click on the game from the steam library and select properties. Then click the button "set launch options". Type "-game Unofficial_Patch" without quotations and you should be all set.
And unless you're skilled with mouse and keyboard I highly suggest using a controller, which was fairly straight forward with steam (I'm using a 3rd party xbox controller).
If you want all a lot of new content. There is clan quest mod. 4.1 came out recently.
It already comes with a version of the Unofficial Patch. 9.2 to be exact. But there is some backported content from the newer releases of the Unofficial Patch. But not all the newer features.

That's very strange and does sound like a bug, yes.
I don't know offhand if the GOG DL includes the basic patch (both the basic and plus patch are by Wes5, https://www.moddb.com/mods/vtmb-unofficial-patch there), but if you don't have it I recommend getting it and restarting your game before you get too deep into it. Plus patch adds to the gaming experience by moving NPCs and altering some dialogue, basic patch just ensures a minimum of game-breaking bugs and crashes happen.
I am waiting for the CQM to be updated to a newer version of the Unofficial Patch and X20 mod.

It's been a long while since I played but I think the mod you need (or at least the one to be able to play the game) is the unofficial patch. That may fix the resolution or there may be another mod for that. Your best bet is to google for what mods are needed.

If you go to the "compatibility" you can find The Darkness in the "Ingame" category which means that the game can be run but experience game-breaking glitches
Your pre-installed UP 10.0 shouldn't matter, as the UP puts its files into a separate folder. (At least that's how it worked for me with the GOG version, which has an older UP installed by default. If in doubt, do a clean reinstall.)
To install the Clan Quest Mod:
You don't have to reinstall it really, you could simply start a fresh game once you're done. I suggest trying the newest patch maybe first, that way you can have the option to skip the sewers.
If you want to play with the images yourself, here's Nines and here's Jack. You can edit the genes to change their appearence. Jack looks a bit haggard, but every time I tried to change that it ended up looking less "right".
I've had fun making these characters but I'm gonna take a bit of a break for now. People have liked the posts so far, but I don't want to spam the subreddit.
Note: Mac OSX tutorial only. Though most steps may be similar for users of other systems.
Step 1: Visit http://debofnight.andcuriouser.com/ and disable the audio player and bottom-right text. > 1.1: Do this by launching Google Chrome and right-click and select the "Inspect" option in the drop-down menu.

Step 2: Open up the website in Full-Screen Mode and Record Your Screen using the application of your choice. > 2.1: You can easily do this with the built-in program Quicktime Player.
Step 3: Save your Screen Recording somewhere (obviously).
Step 4: Download VLC Media Player and Enable "Wallpaper Mode" and Disable "Show Media Title on Video."

> 4.2: Select "Show All" in the bottom-left corner, and under the "Video" tab, you'll find "Enable Wallpaper Mode".

Please note that depending on your Mac OSX, this can take up a lot of CPU (on my Macbook Pro 2012, it takes up about 14-19% of the CPU).
But lucky for us, the newer versions of VLC have a little widget in the menu bar, that allows you to pause the video at any time if your PC starts to overheat because of this.
It really is more of a party-trick though, so don't expect to keep it running 24/7.
Download 7zip if you don't already have it.
Download both of those mods; open both of them up with 7zip (right click on the .zip files, select "open with..." and find 7zip).
Go to the game's main directory (if you need help finding it: here's a guide for Steam)
There will be a folder named "Unofficial_Patch" in the main directory. Go inside of that folder.
With the .zip files for the mods open, drag and drop the contents (or just extract, easy either way) into the "Unofficial_Patch" folder.
If it asks if you want to replace the files, select "yes to all".
Ignore the "Vampire" folder; that's only for the base game without Wesp's patch and any mod put into that folder will not be applied to the Unofficial Patch. These two mods are simple, you just shove the contents into a folder.
Granted these are for the manual downloads option for NexusMods; I have never tried using that other thing (Vortex, I think).
Also, there are some skin mods (mostly ones that were uploaded years ago) that are annoying as fuck to mod cuz you have to rename a bunch of files, but from experience Marius' skin mods are not like that.
I tried it with some random email websites and it didn't work. https://temp-mail.org/en/ was the one suggested to me earlier and it works. I send from Firefox, and open in Chrome Incognito. Make sure you write down the email/don't close the browser window as you'll need it again.
The recent Kindred: The Embraced DVD set has both our remastered Book of Nod, and a newly-filmed epilogue scene with Jeff Kober reprising his role as Daedalus. It's not a full cliff hanger resolution but it helps bring a bit more closure.
